Please start each answer with a new answer book and clearly indicate the number on the front. Section A 1. Your supervisor has provided you with the sequence of a human gene. Explain how you would obtain a suitable template for PCR, and describe in detail the steps you would take to clone this PCR product into an E. coli recombinant protein expression plasmid. Explain how you would validate that the expected clone is correct. Illustrate your answer with diagrams wherever possible. 2. Answer either (a) Describe the typical strategy of determining a three dimensional structure of a macromolecule using X-ray crystallography. Or (b) Describe the application of NMR spectroscopy to study the structure and affinity of a protein-protein complex. Use the following headings as a guide: • Isotope enrichment • HSQC spectrum • Chemical shift perturbation mapping • Binding affinity determination 3. Explain how new technologies such as RNAi, bioinformatics and systems biology enable genome-wide investigations of molecular mechanisms in the cell. Illustrate your answer with diagrams wherever possible. 4. Explain what is meant by the term ‘next-generation sequencing’. Illustrate your answer by describing two next-generation sequencing technologies in detail.
