Section A: Answer TWO questions.

  1. Describe how a bone specimen may be processed for light microscopy. Include in your answer an example of one histological stain that could be used to examine the specimen.
  2. Describe how you would prepare a tissue specimen (choose one specific tissue) for scanning electron microscopy OR transmission electron microscopy. Compare the data you would obtain with your chosen method, with that you would obtain using light microscopy with routine histological staining of the same tissue.
  3. Describe how you would examine the expression of osteopontin in mouse bone sections (as prepared for light microscopy) using enzyme immunochemistry. Include in your answer the necessary precautions and controls to maximize the chances of a successful experiment.
