Using what you’ve learned about restriction enzymes (RE) answer the following questions to determine
the identity of unknown primers used for a PCR reaction with Lambda DNA ( DNA). Below is the map
of DNA and the cut sites for the (RE) EcoRI and HindIII along with the standard DNA ladder
information, these will be helpful in solving this puzzle.
The table below shows three primers for PCR targeting specific DNA regions. For each primer set,
region” refers to the numbered base pair areas of DNA (total of 48,502 linear bp) that are
copied using the primer target sequences.
1. Given the information above, and referring to the DNA map, calculate the length of the PCR product
for each primer set in base pairs (bp) and the size (bp) of the fragment(s) when cut with EcoRI or HindIII.
