Common Initial Measurements

  • pH
  • Titratible Acidity (TA)
  • Malic acid
  • VA

Chemistry

  • Transforms malic acid into lactic acid
  • Lactic acid is less acidic than malic acid
  • May see a reduction in TA between 1 – 3 g/L
  • MLF adds other metabolic products that may contribute to complexity such as diacetyl
  • MLF can be inoculated or may start spontaneously
  • Presence of Lactic acid will show history of MLF activity

Common monitoring methods

  • Visual observation  bubbles in the fermentation lock (good to 100mg/l)
  • Paper chromatography  qualitative method
  • Enzymatic  Precise method
  • HPLC

Malic by Paper Chromatography

  1. placing a small dot or line of sample solution onto a strip of chromatography paper.
  2. The paper is placed in a jar containing a shallow layer of solvent and sealed.
  3. solvent rises through the paper, meets the sample mixture which starts to travel up the paper with the solvent.
